Method

Peeled epithelia are mounted on a recording platform and then are examined under a microscope equipped with a 60× water-immersion objective. Recording electrodes are attached to the basolateral membranes of single TBCs. In these voltage-clamp experiments, voltage-gated currents are amplified and filtered at 10 kHz using a voltage-clamp amplifier. Data are digitized using an A/D converter and are then stored using pCLAMP data acquisition and analysis software on a personal computer. The recording electrodes are filled with an intracellular CsCl solution using a pipette with a resistance of approximately 5 MΩ in physiological saline. To immunohistochemically identify cell types, biocytin (2 mg/ml) is injected through the recording electrodes.
