Description ATP (Adenosine-Triphosphate, Adenosine 5'-triphosphate) is a multifunctional nucleoside triphosphate and an important endogenous signaling molecule in immunity and inflammation.

ATP, a multifunctional nucleoside triphosphate and an important endogenous signaling molecule, restores signaling responses to anti-TCR treatment in Jurkat cells.[2]

Method

Cells are washed with calcium-free and magnesium-free 1×DPBS to remove calcium and magnesium. Then cells are suspended with 450 µL 1×DPBS into a 1.5 mL centrifugal tube, treated with 50 mL Streptolysin O (0.5 µg/ml), and incubated 60 mins at 37℃. Next, cells are centrifuged at 400 g for 4 mins to discard the supernatant. Cells are re-suspended with 100 mL 1×DPBS, treated with 11 µL 50 mM ATP, and incubated in the 37℃ for indicated time.
